I am a graduate student in Tonks lab at Cold Spring Harbor Laboratory. The main focus of our lab is to study protein tyrosine phosphatases (PTPs), which remove phosphate group from their substrates. Protein phosphorylation is one of the major cellular regulatory mechanisms, and the aberrant regulation of phosphorylation status could lead to a variety of diseases, such as metabolic disorders, inflammation, cancers, and so on. My thesis project is to identify and characterize PTPs that contribute to acute myeloid leukemia (AML) pathogenesis. Using CRISPR-Cas9 based negative screen that targeted all the mouse PTPs, I identified Ptpn23 is required for AML cell survival. The future work will focus on mechanism study and evaluation of PTPN23 as a potential drug target to treat AML patients. With the clinical mentorship of Dr. Wadie Bahou, I am eager to explore both the clinical and public health aspects of blood disorders.
